According to Mai (2012: 155–168), these two paths have rendered the cooperative interactions between the traditional and new media outlets far less frequent, if not totally suspended, and are likely to further alienate the newspapers from the original or latest new media departments. In other words, both paths create new administrative barriers whilst breaching or circumventing the original administrative barriers.

Studies in patients and animal and cell models have linked neuronal hyperexcitability to ALS (6, 27–32), but it remains unclear if there are common cell-autonomous mechanisms that drive hyperexcitability in different ALS patients. We previously found that reduced C9ORF72 levels lead to elevated surface-bound levels of glutamate receptors in C9ORF72 ALS iMNs, C9ORF72-deficient mice, and C9ORF72 patients, and that these differences were not caused by differential gene expression (4). We also showed that the elevated glutamate receptor levels increased sensitivity to glutamate-induced excitotoxicity in vitro and in vivo (4).

The definition of full-media convergence is similar to that of media convergence that was developed in light of current practices in Europe and the US. However, as Menke et al. (2016) argued in their comparative study of European convergence journalism, the processes of convergence depend on national and cultural factors, thereby resulting in different convergence practices within specific newsrooms. To gain an improved understanding of the current convergence in China, the factors that make China’s case substantially different from Europe and the US must be considered.

Indeed, we find that C9ORF72 and these sporadic ALS iMNs share defects in autophagosome formation and the aberrant accumulation of glutamate receptors. We find that an engineered, anticoagulation-deficient form of activated protein C called 3K3A-APC, but not heat-inactivated 3K3A-APC (inactive 3K3A-APC), can restore autophagosome formation in both C9ORF72 ALS/FTD and sporadic ALS iMNs. Interestingly, 3K3A-APC also normalizes glutamate receptor levels in C9ORF72 and sporadic ALS iMNs. As a result of these activities, 3K3A-APC lowers dipeptide-repeat protein (DPR) levels in C9ORF72 ALS/FTD iMNs, and potently restores nuclear TDP-43 localization and the normal survival of both C9ORF72 and sporadic ALS iMNs. We show that the ability of 3K3A-APC to rescue ALS iMN survival is dependent on its ability to activate protease-activated receptor 1 (PAR1), identifying PAR1 as a therapeutic target for both C9ORF72 and sporadic ALS. Importantly, 3K3A-APC also rescues glutamate receptor levels and proteostasis impairments in vivo. Thus, motor neurons from C9ORF72 and at least a subset of sporadic ALS patients share common defects in autophagosome formation and glutamate receptor homeostasis, and pharmacologic rescue of these defects by PAR1 activation may slow or prevent neurodegeneration in a substantial fraction of ALS cases.

Evidently, these questions can be explored in future research.

Total RNA was extracted from sorted iMNs on day 21 after transduction with a TRIzol RNA Extraction Kit (Life Technologies) and reverse transcribed with an Oligo dT primer using a ProtoScript II First Strand Synthesis Kit (NEB). From the viewpoint of local journalists, news from the Fujian press industry was originally highly homogenised and lacked competitiveness. After the three-phase censorship of new media was institutionalised, risks from converging news production declined, but the timely release of information was weakened and the opportunity for journalistic autonomy has decreased as well. When covering politically sensitive social events, the freedom of speech of the new media was equally limited relative to others within the press industry and was occasionally even more restricted than that of newspapers.
Before and after 2009, all major press groups in Fujian started to establish e-newspapers and transfer the newspaper contents to their official news websites to cope with the loss of readers as a result of the prevalence of computer technology as well as to maintain and expand the influence of traditional media. The popularity of Weibo and WeChat emerged between 2011 and 2012, thereby prompting the local press groups in Fujian to try out both platforms. During this period, newspapers were the pillar of the press groups’ revenue. Within most press groups in Fujian, Weibo and WeChat remained under the operation of newspaper editors and journalists on a part-time basis and received limited emphasis from the leadership.

Our data indicate that motor neurons derived from C9ORF72 and at least a subset of sporadic ALS patients share similar defects in autophagosome formation and glutamate receptor accumulation. Importantly, PAR1 activation by 3K3A-APC can reverse these C9ORF72 and sporadic ALS disease processes in vitro and in vivo (Figure 7). We draw several key conclusions from our findings.
Moreover, the major criteria of the present press industry in Fujian regarding job performance of new media editors involve the read count of postings and the number of fans. From the perspective of journalists, behaviour that editors repeatedly demand from the frontline journalists includes nothing more than ensuring the timeliness of press release. Their compliance is somehow related to the job performance evaluation of the editors themselves, thereby resulting in the additional decline in the willingness for coordination.

The differences in the reactions to the encouragement between young/junior and old/senior journalists follow the same logic. Unlike young and junior journalists who hold contract positions, the old and senior journalists have substantial security from their authorised status (Bianzhi), thus rarely suffer from the effects of the living pressure of newspapers regarding current journalistic practices.

The relationship between demographic factors with journalists’ attitude towards new media has been analysed in order to determine the variation among different types of journalists. The results of the ANOVA test indicate that gender and degree of education make no difference to the respondents’ perception about the changes or to their evaluation of new media; the type of newspaper makes no difference to the former but does influences the latter (F = 13.107, df = 1, p < .01). The results of the correlation analysis indicate that the respondents’ ages and years at work are uncorrelated to their perception but positively correlated to their evaluation of new media (r = 0.235, p < .001; r = 0.185, p < .01). Although local journalists in Fujian have generally recognised the changes brought about by new media to newspapers, those from metropolis newspapers were more inclined to provide a negative evaluation to these changes than those from party organs, whilst the older and more experienced the journalists were, the more positive evaluation they tended to provide to new media.

Simply speaking, media convergence is the process of integrating new media into the system of traditional media. Thus, journalists’ attitude towards the new media constitutes the foundation for better understanding of journalists’ attitude towards the convergence between traditional media and new media. Moreover, media convergence carries a prognosis that news production will move further to the direction towards timeliness, interactivity and use-centredness. Given that the newsroom routine adjusts rapidly, particularly as the audience increasingly engages in news production and professional boundaries blur further, the following questions need to be answered: Will the viewpoint of journalists towards new media become considerably negative? How will this attitude affect their perception of the convergence process between new media and traditional media?
Fujian’s case indicates that new media outlets tend to be ‘domesticated’ in terms of converging news production. Firstly, a multilevel censorship system has been established, thereby ensuring that the new media content is under supervision. Secondly, new media outlets are subject to a bureaucratic style of management with low decision-making efficiency.

Moreover, the decline of influence on public opinion is definitely another key factor that triggered the convergence within Chinese press industry. It is fairly illustrated in Secretary-General Xi Jinping’s ‘8•19’ speech on ‘boost the convergence between traditional and new media, and completely apply new technology in innovating the forms of media communication to seize a commanding height in the field of information communication’ (Liu, 2014). This situation is relatively similar to that of the media conglomeration in the late 1990s, when the legitimacy of reform stemmed from the policy of making domestic media ‘bigger and stronger’ to pre-empt the anticipated foreign competition in the World Trade Organization era. Meanwhile, a majority of domestic press groups were established based on administrative decrees rather than on business demands (Chen and Lee, 1998). Eliminating ‘dispersion’ and ‘chaos’ in the public opinion domain is the key factor that catalyses both media conglomeration and media convergence.

In the early 17th century, persistent drought and famine driven by the Little Ice Age accelerated the collapse of the Ming dynasty.[9] Two major popular uprisings swelled up, led by Zhang Xianzhong and Li Zicheng, both poor men from famine-hit Shaanxi who took up arms in the 1620s.[10] At the same time, Ming armies were occupied in the defence of the northern border against the Manchu ruler Hong Taiji, whose father, Nurhaci, had united the Manchu tribes into a cohesive force. 3K3A-APC potently rescued autophagosome formation in C9ORF72 ALS iMNs, as determined by its ability to increase the number of GFP+mRFP+ vesicles in bafilomycin-treated iMNs (Figure 2, A and B [C9-ALS, inactive 3K3A-APC, +bafilomycin vs. C9-ALS, 3K3A-APC, +bafilomycin]; 3 controls and 3 C9ORF72 ALS patients). Interestingly, 3K3A-APC also increased autophagosome formation in sporadic ALS iMNs (Figure 2, A and C [sporadic ALS, inactive 3K3A-APC, +bafilomycin vs. sporadic ALS, 3K3A-APC, +bafilomycin]; 3 controls and 5 sporadic ALS patients, and Supplemental Figure 2B), indicating that is also capable of rescuing C9ORF72-independent autophagy impairments. Western blot analysis of C9ORF72 ALS motor neurons showed that 3K3A-APC increased the ratio of LC3-II to LC3-I in the presence of bafilomycin, verifying the results of the mRFP-GFP-LC3 assay (Supplemental Figure 2, C and D). Therefore, 3K3A-APC treatment rescues autophagosome formation in C9ORF72 and sporadic ALS iMNs.
