iPSCs were tested for mycoplasma before, during, and after the study and were negative. iPSCs were first differentiated into fibroblast-like cells to enable efficient retroviral transduction. iPSCs (1 × 106 cells/flask) were seeded in a T75 flask that was coated with Matrigel (Corning) and cultured in mTeSR until reaching 70% to 80% confluence. Cells were then cultured in fibroblast medium (20% FBS in DMEM) for 14 days. Cells were passaged 1:1 with Accutase (Innovative Cell Technologies) to another flask that was coated with Matrigel and cultured in fibroblast medium until they reached 90% confluence. Cells were then passed with 0.25% trypsin to 3 flasks that were coated with 0.1% gelatin and cultured in 10% FBS in DMEM. Reprogramming of the fibroblast-like cells was performed in 96-well plates (8 × 103 cells/well) or 13-mm plastic coverslips (3.2 × 104 cells/coverslip) that had been sequentially coated with gelatin (0.1%, 1 hour) and laminin (2 to 4 hours) at room temperature. Seven iMN factors were added in 100 to 200 μL fibroblast medium per well of the 96-well plate with 5 mg/mL polybrene. Cultures were transduced with lentivirus encoding the Hb9::RFP reporter 48 hours after transduction with transcription factor–encoding retroviruses. On day 5, primary mouse cortical glial cells from P1 ICR pups were added to the transduced cultures in glia medium containing MEM (Life Technologies), 10% donor equine serum (HyClone), 20% glucose (Sigma-Aldrich), and 1% penicillin/streptomycin. On day 6, cultures were switched to N3 medium containing DMEM/F12 (Life Technologies), 2% FBS, 1% penicillin/streptomycin, N2 and B27 supplements (Life Technologies), 7.5 μM RepSox (Selleck), and 10 ng/mL each of GDNF, BDNF, and CNTF (R&D Systems). The iMN cultures were maintained in N3 medium, changed every other day, unless otherwise noted.
C9ORF72 regulates autophagy by promoting the RAB1A-dependent trafficking of the ULK1 autophagy initiation complex to the phagophore (16). Consequently, mTOR inhibitors were shown to be ineffective at rescuing autophagosome formation in C9ORF72-deficient neurons and it is unclear if pharmacological agents can restore normal autophagy initiation (16).
Meanwhile, partisans of the Donglin Academy faction, which had been devastated under Wei Zhongxian's influence, established political organizations throughout the Jiangnan region.[5] Chief among these was the Fushe, or Restoration Society, whose members were a new generation of scholars who identified with the old Donglin faction.[6] They succeeded in placing their members into high government posts through the imperial examinations of 1630 and 1631. The reversal of Wei Zhongxian's fortunes resulted in a renewal of the Donglin faction's influence at court, arousing great suspicion from the Chongzhen Emperor.[7] The nomination of Donglin favorite Qian Qianyi for the post of Grand Secretary led to accusations of corruption and factionalism by his rival Wen Tiren. Qian Qianyi was imprisoned on the emperor's orders. Though he was soon released, his status was reduced to that of a commoner and he returned to Jiangnan. Wen Tiren would later become Grand Secretary himself.[8]

This article introduces child-raising blogs in China. Written by mothers and their 6- to 12-year-old offspring, child-raising blogs contain diverse material from and about a child’s daily life and issues relating to a mother raising her offspring. Six blogs from mothers and their daughters are studied under the aspect of generational relations and the voice of children online with the aim of understanding whether or how child-raising blogs in China are part of a changing discourse about the mother-daughter relationship. The article finds that as the objectified state of cultural capital, blogs present a smoothed-out hierarchical relationship between mother and daughter, and carry the possibility of bestowing a voice on children. Due to their continuous and permanent structure, which records opinions and content deemed important by the authors, blogs could be a catalyst or at least a magnifying lens for changes that are already occurring in family relations offline.

C9ORF72 ALS iMNs have fewer lysosomes than control iMNs (4), which could affect autophagic flux downstream of autophagosome formation. To determine if 3K3A-APC affects lysosome number in C9ORF72 ALS iMNs, we examined the number of LAMP2+ vesicles in iMNs (4). 3K3A-APC restored normal lysosome numbers in C9ORF72 ALS iMNs (Figure 2E; 2 controls and 2 C9ORF72 ALS patients, and Supplemental Figure 2, L and M). 3K3A-APC did not alter C9ORF72 protein (Supplemental Figure 2, N and O; 3 C9ORF72 ALS patients) or mRNA levels (Supplemental Figure 2P) in C9ORF72 ALS iMNs. Therefore, 3K3A-APC rescues lysosome numbers in C9ORF72 ALS iMNs without altering C9ORF72 levels.

These findings provide an empirical foundation to organise the interview outline and the type of interviewees that should be recruited in the following interviews. More importantly, the results mentioned above indicate that journalists in Fujian do embrace new technologies. In this case, the journalists are not resisting media convergence because they have negative attitudes towards new technologies but due to institutional, organisational and value reasons, which will be discussed thoroughly in the next section.

From the viewpoint of local journalists, news from the Fujian press industry was originally highly homogenised and lacked competitiveness. After the three-phase censorship of new media was institutionalised, risks from converging news production declined, but the timely release of information was weakened and the opportunity for journalistic autonomy has decreased as well. When covering politically sensitive social events, the freedom of speech of the new media was equally limited relative to others within the press industry and was occasionally even more restricted than that of newspapers.
One of the goals of media convergence in the reconstruction process is to modify the journalist–editor relationship by veering from ‘journalist-centred’ to ‘editor-centred’, such that journalists shall mainly undertake the role of information gatherer, whereas editors perform the role of information synthesiser. Regarding real news production, most new media centre editors are recruited from the members of society who are familiar with new media but inexperienced in news gathering and editing; thus, they are mere ‘stevedores’ rather than ‘processors’ of texts. Given that editors lacked the required qualifications, journalists were forced to undertake additional tasks (e.g., verifying information and converting statements from frontline journalists into text) that should have been performed by editors.
Therefore, faced with the challenges from we-media runners who have seized advantages in the information communication domain in recent years, local journalists choose to fall back on the principle of objectivity to defend the increasingly blurring boundary between professional and non-professional information providers. The resistance to collaboration thereby demonstrates the journalists’ identification with the ideology of professionalism.

In 1645, Zhu Yousong, who had proclaimed himself the Hongguang Emperor of the Southern Ming dynasty, gave the Chongzhen Emperor the temple name "Sizong". In historical texts, "Sizong" is the most common temple name of the Chongzhen Emperor, even though the Southern Ming rulers had changed "Sizong" to "Yizong" (毅宗) and then to "Weizong" (威宗). During the Qing dynasty, the Chongzhen Emperor's temple name was changed to "Huaizong" (懷宗).
Amyotrophic lateral sclerosis (ALS) is a fatal motor neuron disease with diverse etiologies. Therefore, the identification of common disease mechanisms and therapeutics targeting these mechanisms could dramatically improve clinical outcomes. To this end, we developed induced motor neuron (iMN) models from C9ORF72 and sporadic ALS patients to identify targets that are effective against these types of cases, which together comprise approximately 90% of patients. We find that iMNs from C9ORF72 and several sporadic ALS patients share 2 common defects — impaired autophagosome formation and the aberrant accumulation of glutamate receptors. Moreover, we show that an anticoagulation-deficient form of activated protein C, 3K3A-APC, rescues these defects in both C9ORF72 and sporadic ALS iMNs. As a result, 3K3A-APC treatment lowers C9ORF72 dipeptide-repeat protein (DPR) levels, restores nuclear TDP-43 localization, and rescues the survival of both C9ORF72 and sporadic ALS iMNs. Importantly, 3K3A-APC also lowers glutamate receptor levels and rescues proteostasis in vivo in C9ORF72 gain- and loss-of-function mouse models. Thus, motor neurons from C9ORF72 and at least a subset of sporadic ALS patients share early defects in autophagosome formation and glutamate receptor homeostasis and a single therapeutic approach may be efficacious against these disease processes.

Males and females accounted for 49.6% and 50.4%, respectively, of the total number of respondents. The respondents with a degree of and below junior college accounted for 1.8%, those with bachelor’s degree accounted for 83.5% and those with a master’s degree or above accounted for 14.7%. Among the respondents, 33.2% came from party organs, whilst 66.8% came from metropolis newspapers. The mean age of the respondents was 31 years old (M = 30.63, SD = 6.35) and the mean number of years at work was 8 (M = 7.62, SD = 6.50).

Last year or the year before, I went to the trial over a deputy mayor in Fuzhou. Only another journalist and I went to the whole course of the first trial. He (a newspaper department supervisor) demanded strictly at that time that the number of online figures could not exceed 500. Plus, the next day he scolded (another journalist) in a loud voice, ‘It’s too foolish of you to have (only) offered them the lead of the foregoing news. You should have cut out a bit from each paragraph. You giving these 500 figures to others, who would read the newspaper?’ (Interviewee No. 14)
Mice were anesthetized with i.p. ketamine (100 mg/kg) and xylazine (10 mg/kg), and body temperature kept at 36.9°C ± 0.1°C with a thermostatic heating pad. Mice were placed in a stereotactic apparatus (ASI Instruments) and the head was fixed accordingly. A burr hole was drilled, and an injection needle (33 gauge) was lowered into the hippocampus between CA1 and the dentate gyrus (anteroposterior −2.0, mediolateral +1.5, dorsoventral −1.8). NMDA (10 nmoles) and 0.2 μg of 3K3A-APC or vehicle in 0.3 μL of PBS, pH 7.4 was infused over 2 minutes using a micro-injection system (World Precision Instruments).
Our findings provide preclinical evidence suggesting that 3K3A-APC and PAR1 activation could rescue neurodegeneration in C9ORF72 and sporadic ALS cases. In complex diseases such as ALS, the identification of therapeutic strategies that are efficacious across a wide range of cases is critical for clinical success. We have developed tools, including C9ORF72 and sporadic ALS iMN disease models, that enable the identification of these rare, but critical therapeutic strategies.
In the past when our Weibo and WeChat accounts were not taken over (by the new media centre of Xiamen Daily)… Journalists running to the spot would make an instant call to our editors, saying how a piece was on WeChat… Nowadays, the journalists no longer have such enthusiasm… Anyway, whatever the achievement is, they are not ours. We hypothesized that phenotypic analysis of iMNs from these sporadic ALS patients and C9ORF72 ALS/frontotemporal dementia (FTD) patients would enable the identification of degenerative mechanisms that are shared by C9ORF72 and at least a subset of sporadic ALS motor neurons. We further hypothesized that pharmacologic agents capable of normalizing these phenotypes in C9ORF72 and sporadic ALS iMNs might also rescue neurodegeneration and lead to the identification of therapeutic targets with broad efficacy in the ALS population.
