In April 1644, the Ming imperial court finally ordered Wu Sangui to move his army south from his fortress at Ningyuan to Shanhai Pass.[17] It was too late, however, and Wu would not reach Shanhai Pass until 26 April.[18] Word reached Beijing that Shun rebels were approaching the capital through Juyong Pass, and the Chongzhen Emperor held his last audience with his ministers on 23 April. Li Zicheng offered the emperor an opportunity to surrender, but the negotiations produced no result. Li commanded his forces to attack on 24 April. Rather than face capture by the rebels, the Chongzhen Emperor gathered all members of the imperial household except his sons. Using his sword, he killed Consort Yuan and Princess Zhaoren, and severed the arm of Princess Changping.
Focusing on the ‘top-down design’ of media convergence, market-oriented research relatively lacks in social solicitude and theoretical construction; valuing the ethics and obligations of converging media, the norm-oriented research demonstrates intense social solicitude but hardly receives industry-wide recognition. Both types of research fail to explain the full influence of media convergence. Considering that journalists are the critical actors of news production, the advancement of media convergence cannot proceed without the institutional arrangement from the top to the bottom as well as without the commitment and involvement of journalistic practitioners, particularly journalists, from the bottom to the top. The interaction between these two processes ultimately determines the scope, intensity and nature of the influences of media convergence.

To determine if the effects of 3K3A-APC on stimulating autophagy in C9ORF72 and sporadic ALS iMNs were sufficient to lower DPR and cytoplasmic TDP-43 levels, we first measured levels of glycine-arginine [poly(GR)] and proline-arginine [poly(PR)] DPRs in C9ORF72 ALS iMNs with and without 3K3A-APC treatment. Treatment with 3K3A-APC for 6 days reduced the number of nuclear poly(GR)+ and poly(PR)+ punctae in C9ORF72 ALS iMNs to control levels (Figure 3, A–C, and Supplemental Figure 3, A–E; 2 controls and 2 C9ORF72 ALS patients). Dot blot analysis for poly(GR) confirmed the decrease in iMNs after 3K3A-APC treatment (Figure 3, D and E). Moreover, 3K3A-APC reduced total nuclear and cytoplasmic levels of poly(GR)+ and poly(PR)+ punctae in C9ORF72 ALS iMNs (Supplemental Figure 3, F–H and J–L) and did not significantly alter the ratio of nuclear to cytoplasmic poly(PR)+ punctae, although it slightly lowered the ratio of nuclear to cytoplasmic poly(GR)+ punctae intensity (Supplemental Figure 3, I and M). These data indicate that 3K3A-APC treatment decreased both nuclear and cytoplasmic levels of DPRs. qRT-PCR using primers specific for C9ORF72 variants 1 and 3 (Supplemental Table 3) showed that 3K3A-APC did not reduce RNA levels of the repeat expansion (Supplemental Figure 3N), excluding the possibility that the reduced DPR levels were caused by transcriptional repression. 3K3A-APC also did not alter the expression of genes associated with the integrated stress response (Supplemental Figure 3O), indicating that its ability to lower DPR levels was independent of this pathway.

The effects of the dominant path of media convergence on the mid-size press industry are the focus of this study. The path chosen by the press industry in other regions of China may differ from the dominant one in Fujian due to the nuances of power structure, level of economic development, size of press group and other factors. For example, the Nanfang Media Group’s choice of the fully transformed path has a bearing on its pioneering spirit and remarkable investment, which are derived from its distance from the centre of political power, the prosperity of the market economy in Guangdong and the surrounding areas, and the considerable assets from accumulation within the media group in the past decades (Yin and Liu, 2013). Nevertheless, Fujian’s case discloses the common challenges that the Chinese press industry will face as they further advance the convergence regardless of which path they choose. This study also develops an analytical framework based on the interplay amongst the state, media and journalists to be used in future research on China’s media convergence.
Moreover, the decline of influence on public opinion is definitely another key factor that triggered the convergence within Chinese press industry. It is fairly illustrated in Secretary-General Xi Jinping’s ‘8•19’ speech on ‘boost the convergence between traditional and new media, and completely apply new technology in innovating the forms of media communication to seize a commanding height in the field of information communication’ (Liu, 2014). This situation is relatively similar to that of the media conglomeration in the late 1990s, when the legitimacy of reform stemmed from the policy of making domestic media ‘bigger and stronger’ to pre-empt the anticipated foreign competition in the World Trade Organization era. Meanwhile, a majority of domestic press groups were established based on administrative decrees rather than on business demands (Chen and Lee, 1998). Eliminating ‘dispersion’ and ‘chaos’ in the public opinion domain is the key factor that catalyses both media conglomeration and media convergence.

In this work we push forward parameter identifiability of linear mixture models (LMM) to nonlinear ones. LMM finds many applications in blind source separation-related problems, e.g., hyperspectral unmixing and topic mining. In practice, however, the mixing process is hardly linear. This work studies a fundamental question: if there is nonlinearity imposed upon an LMM, can we still identify the underlying parameters of interest? The interesting observation of our work is that: under some conditions, nonlinearity can be effectively removed and the problem will boil down to an LMM identification problem — for which we have tons of tools to handle.

To date, the pioneers of media convergence have been thoroughly studied by Chinese scholars. Other press groups as ‘followers’, particularly the local press groups that adopted the dominant path of media convergence, are less highlighted and rarely focused within journalism studies, although they are precisely what have crucially endowed the landscape of Chinese media convergence with regional diversity. The deficiency in relevant studies has entailed the urgent choice of Chinese scholars to focus on media convergence of the local press industry.

To determine how 3K3A-APC might stimulate autophagosome formation, we performed RNA sequencing (RNA-Seq) on C9ORF72 ALS iMNs treated with 3K3A-APC or inactive 3K3A-APC for 3 days (2 C9ORF72 ALS patients). Consistent with our findings in the mRFP-GFP-LC3 and immunoblot assays, both Ingenuity Pathway Analysis (IPA) and KEGG pathway analysis revealed “autophagy” as one of the most significantly enriched gene sets (Supplemental Figure 2, E–H). RNA-Seq and quantitative real-time PCR (qRT-PCR) showed that 3K3A-APC significantly increased the expression of ATG5 and ATG10, which encode proteins with key roles in driving autophagosome formation (ref. 25, Figure 2D [qRT-PCR], Supplemental Figure 2I [RNA-Seq], and Supplemental Table 4). Thus, 3K3A-APC increases the expression of genes encoding autophagy-initiating factors in ALS iMNs.

Whilst ruling his kingdom, Goujian never relished kingly riches, but instead ate food suited for peasants, as well as forcing himself to taste bile, in order to remember his humiliations while serving under the State of Wu. The second half of a Chinese idiom, wòxīn-chángdǎn (臥薪嚐膽, "sleeping on sticks and tasting gall"), refers to Goujian's perseverance.
Both viewpoints have consistent cores, that is, the adherence to the principle that ‘content shall dominate’, which underscores the importance of content resources in maintaining and promoting the influential power of newspapers and in assisting newspapers to step out of the ‘cold winter’. This situation further evokes the hesitation of and the resistance from the journalists of the Fujian press industry as they maintain their professional dignity with effort.
