For all other experiments, analyses of iMN samples were performed with the statistical software package Prism Origin v.7.0a (GraphPad Software). The normal distribution of data sets was tested by the D’Agostino-Pearson omnibus normality test. Differences between multiple groups were analyzed using 1-way ANOVA with Tukey’s correction for all comparisons, unless the data were non–normally distributed for which nonparametric Kruskal-Wallis testing was used. Mean and standard deviation or standard error of the mean was used for normally distributed data sets, and the median and interquartile range were used for non–normally distributed data sets. Differences between 2 groups were analyzed using a 2-tailed Student’s t test, unless the data were non–normally distributed, in which case 2-sided Mann-Whitney testing was used. Significance was assumed at P < 0.05.

It is further revealed in this study that the newsroom routines, multiskilling and other factors that significantly affect the attitude of journalists in European and American countries towards media convergence do not sufficiently account for the reluctance of Chinese journalists. The latter is ascribed to the institutional, organisational and individual complexities of the Chinese press industry, within which the competition for market between the traditional and new media departments is implicitly permitted as long as it does not threaten the bottom line of political safety, and the ideology of professionalism with which the journalists identify provides the justification for their willingness to confront the severe challenges from the non-professional information providers (e.g., we-media) by collaborating with the new media centre.
By this point, the situation had become critical for the Chongzhen Emperor, who rejected proposals to recruit new militias from the Beijing region and to recall general Wu Sangui, the defender of Shanhai Pass on the Great Wall. The Chongzhen Emperor had dispatched a new field commander, Yu Yinggui, who failed to stop Li Zicheng's armies as they crossed the Yellow River in December 1643. Back in Beijing, the capital defence forces consisted of old and feeble men, who were starving because of the corruption of eunuchs responsible for provisioning their supplies. The troops had not been paid for nearly a year.[15] Meanwhile, the capture of Taiyuan by Li Zicheng's forces gave his campaign additional momentum; garrisons began to surrender to him without a fight. Through February and March 1644, the Chongzhen Emperor declined repeated proposals to move the court south to Nanjing, and in early April, he rejected a suggestion to move the crown prince to the south.[16]

Because autophagy induction can enhance TDP-43 turnover in motor neurons (26), we wondered whether 3K3A-APC treatment could prevent the cytoplasmic accumulation of TDP-43 in ALS iMNs. 3K3A-APC treatment for 6 days significantly increased the nuclear-to-cytoplasmic ratio of TDP-43 in C9ORF72 and sporadic ALS iMNs to control levels (Figure 3, G–J; 2 controls, 2 C9ORF72 ALS, and 6 sporadic ALS patients). Thus, 3K3A-APC treatment can rescue autophagosome formation, lower DPR levels in C9ORF72 ALS iMNs and rescue DPR-mediated neurotoxicity, and reverse the cytosolic accumulation of TDP-43 in C9ORF72 and sporadic ALS patient iMNs.

The recruitment of interviewees was conducted for two rounds. Given the demonstrative effect of the media convergence of the Xiamen Daily Group within the province and even the entire industry, the authors contacted seven media practitioners from this organisation to undergo the first round of interview. Consequently, one director, one editor and five newspaper journalists were involved as interviewees in this round. After developing a preliminary knowledge of the opinions of the Xiamen Daily Group journalists towards media convergence, the authors conducted a second round of interview. Three directors, three editors and seven journalists were recruited as interviewees from several other press groups, including Fujian Daily Group, Fuzhou Daily News Press and Quanzhou Evening News Press, thereby promoting the diversity of the interviewees in the aspect of age, years at work and newspaper type. Except for one journalist from the Fujian Daily Group who was interviewed via telephone, all the 19 others underwent face-to-face interviews (Table 1).

Zhu Youjian was the fifth son of Zhu Changluo, the Taichang Emperor, and one of his low-ranking concubines, Lady Liu. When Zhu Youjian was four years old, his mother was executed by his father for reasons unknown and was buried secretly. Zhu Youjian was then adopted by his father's other concubines. Previous studies on the influence of media convergence in China either took a market- or norm-oriented approach. From a news production perspective, the current study analyses the interaction between the top-down design and bottom-up practices of journalists to disclose the influence of the dominant path of media convergence within the press industry of Fujian Province. A survey and 20 in-depth interviews show that the current media convergence practices of Fujian’s press industry fail to receive the support of journalists because of institutional, organisational and individual complexities, rather than technological reasons. This study discusses the implications of this finding for media convergence in China.

59 95% confidence interval ranged between 55 and 90%. Old Wang is only 33% likely to read commercialized papers (95% confidence ranged between 24 and 42%). A dummy variable for having travelled to countries in Europe or North America was dropped from the analysis, because it predicted the use of commercialized papers perfectly. Results can be retrieved from the author upon request.

Two alternative paths are presently available for the local press industry to thoroughly address the issue of content for new media outlets. The first path is to remove the administrative hierarchy between metropolis newspapers and party organs, with the new media centre providing news to both newspapers and new media outlets for further editing. In this case, the new media outlets are not incorporated into the new media centre but remain at the same administrative level as the traditional media departments. Hence, the vertical communication between the new media centre and various new media outlets increases, yet the horizontal interaction between newspapers and new media outlets dramatically reduces. The second path is to insulate the newspapers from converging news production, with the centre producing content independently for new media outlets.
A key molecular hallmark of ALS is the nuclear depletion and cytoplasmic accumulation of TDP-43 in motor neurons (14). The cytosolic accumulation of TDP-43 is thought to drive motor neuron degeneration (14, 15). Immunofluorescence verified that both C9ORF72 and sporadic ALS iMNs possessed a lower nuclear-to-cytoplasmic ratio of TDP-43 than controls (Figure 1, G–I; 2 controls, 2 C9ORF72 ALS, and 6 sporadic ALS patients). Thus, these sporadic ALS iMNs exhibit neurodegenerative processes.
As national policy is further loosened, the Chinese media have begun exploring diversified modes of business operation. In 1996, the Guangzhou Daily Newspaper Group announced its foundation, thereby unfolding the prelude of conglomeration of Chinese media. However, the process of conglomeration accelerated significantly only during the turn of the millennium, which has also benefited from the promulgation of a series of polices.

To determine if increased glutamate receptor levels might be a conserved mechanism that drives hyperexcitability in multiple forms of ALS, we examined levels of the NR1 NMDA receptor subunit in neurites of control, C9ORF72 ALS, and sporadic ALS iMNs by immunostaining. Similarly to our previous study, C9ORF72 iMNs possessed significantly more NR1+ punctae on neurites than control iMNs (Figure 4, A and B; 2 controls and 2 C9ORF72 ALS patients, and ref. 4). Colabeling with a MAP2-specific antibody verified that the NR1+ punctae that were increased in abundance were localized on dendrites (Supplemental Figure 4, A and B). Interestingly, sporadic ALS iMNs also displayed more NR1+ punctae on neurites than control iMNs (Figure 4, A and C, and Supplemental Figure 4, C–H; 2 controls and 6 sporadic ALS patients). Calcium imaging confirmed that C9ORF72 and sporadic ALS iMNs experienced more calcium transients than controls in response to glutamate, indicating that they may be more sensitive to excitotoxicity (Figure 4, D and E; 3 controls, 3 C9ORF72 ALS patients, 1 sporadic ALS patient). Thus, both C9ORF72 and sporadic ALS iMNs display increased NR1 levels, which could reflect a shared mechanism of increased susceptibility to excitotoxicity.

Moreover, most journalists willing to provide news to the new media centres are from party organs, whereas the majority of the metropolis newspaper journalists resisted collaboration. To understand the reason behind such contrast, note that new media centres are governed by party organs in terms of administrative hierarchy and are completely independent of metropolis newspaper offices in terms of finances. Compared with the metropolis newspapers, the party organ has relatively few barriers to the new media centres and a high degree of resource sharing. Such disparity has resulted in an operational obstacle between metropolis newspaper offices and the new media centre. To the new media leadership, this obstacle is precisely the underlying reason for newspapers and the new media to continue to be integrated but stay incompatible. The pattern of this relationship affects journalists’ willingness to collaborate.

It is said that even in his lifetime a few of Wang’s characters or his signature were priceless. Down through the ages, aspiring students of that most basic yet highest art in China, calligraphy, have copied and preserved traces of his style. The most famous example of his writing is the Lantingxu (“Preface to the Poems Composed at the Orchid Pavilion”), which recorded a famous gathering of some 42 literary figures during the Spring Purification Festival of 353 ce to compose poems and enjoy the companionship of wine. There are at least 3 potential reasons for the shared abnormalities in autophagosome formation and glutamate receptor levels we have observed. First, some sporadic ALS patients may harbor genetic variants that directly impair autophagosome formation. If so, this could induce the autophagosome formation defects that in turn could cause excess glutamate receptor accumulation. Alternatively, genetic variants affecting other aspects of proteostasis could lead to the accumulation of unfolded proteins, which might impair autophagosome formation through some unknown mechanism. The accumulation of unfolded protein could also impair overall protein turnover and lead to glutamate receptor accumulation. Third, the sporadic ALS patients could harbor genetic variants that affect vesicular signaling molecules, such as phosphatidylinositols, that regulate both endocytosis of surface proteins and autophagosome formation (38). It will also be important to determine how conserved these neuronal phenotypes are in other neurodegenerative diseases. One study has clearly shown that a Parkinson’s disease mutation in synaptojanin impairs autophagosome formation in iPSC neurons (38).
We hypothesized that phenotypic analysis of iMNs from these sporadic ALS patients and C9ORF72 ALS/frontotemporal dementia (FTD) patients would enable the identification of degenerative mechanisms that are shared by C9ORF72 and at least a subset of sporadic ALS motor neurons. We further hypothesized that pharmacologic agents capable of normalizing these phenotypes in C9ORF72 and sporadic ALS iMNs might also rescue neurodegeneration and lead to the identification of therapeutic targets with broad efficacy in the ALS population.
