To determine if the effects of 3K3A-APC on stimulating autophagy in C9ORF72 and sporadic ALS iMNs were sufficient to lower DPR and cytoplasmic TDP-43 levels, we first measured levels of glycine-arginine [poly(GR)] and proline-arginine [poly(PR)] DPRs in C9ORF72 ALS iMNs with and without 3K3A-APC treatment. Treatment with 3K3A-APC for 6 days reduced the number of nuclear poly(GR)+ and poly(PR)+ punctae in C9ORF72 ALS iMNs to control levels (Figure 3, A–C, and Supplemental Figure 3, A–E; 2 controls and 2 C9ORF72 ALS patients). Dot blot analysis for poly(GR) confirmed the decrease in iMNs after 3K3A-APC treatment (Figure 3, D and E). Moreover, 3K3A-APC reduced total nuclear and cytoplasmic levels of poly(GR)+ and poly(PR)+ punctae in C9ORF72 ALS iMNs (Supplemental Figure 3, F–H and J–L) and did not significantly alter the ratio of nuclear to cytoplasmic poly(PR)+ punctae, although it slightly lowered the ratio of nuclear to cytoplasmic poly(GR)+ punctae intensity (Supplemental Figure 3, I and M). These data indicate that 3K3A-APC treatment decreased both nuclear and cytoplasmic levels of DPRs. qRT-PCR using primers specific for C9ORF72 variants 1 and 3 (Supplemental Table 3) showed that 3K3A-APC did not reduce RNA levels of the repeat expansion (Supplemental Figure 3N), excluding the possibility that the reduced DPR levels were caused by transcriptional repression. 3K3A-APC also did not alter the expression of genes associated with the integrated stress response (Supplemental Figure 3O), indicating that its ability to lower DPR levels was independent of this pathway.

To determine if 3K3A-APC can normalize glutamate receptor levels on neurons in vivo, we examined its ability to lower NR1 levels and NMDA-induced excitotoxicity in C9orf72+/– mice. We previously showed that hippocampal, cortical, and spinal cord neurons of C9orf72+/– mice have increased NR1 levels compared with C9orf72+/+ mice (4). We previously developed an acute, in vivo NMDA-induced excitotoxicity assay in the hippocampus (Figure 6E), and verified using cresyl violet staining as well as Fluoro-Jade B–positive dead neuron counting in sham control mice that the operation procedure itself does not result in significant injury (4). Similarly to iMN cultures, reduced C9ORF72 levels in C9orf72+/– and C9orf72–/– mice cause hippocampal neurons to undergo greater NMDA-induced excitotoxicity than in C9orf72+/+ mice, resulting in a significantly larger loss of neurons in C9ORF72-deficient mice that could be detected by a reduction in cresyl violet staining (Figure 6, F and G, and ref. 4). A single injection of 3K3A-APC into the hippocampus of C9orf72+/– mice significantly lowered NR1 levels after 48 hours (Figure 6, H and I) (n = 3 mice). Moreover, when coinjected with NMDA, 3K3A-APC significantly reduced the amount of NMDA-induced hippocampal neurodegeneration in C9orf72+/– mice (Figure 6, F and G). Thus, in vivo, 3K3A-APC improves proteostasis and normalizes glutamate receptors, thereby rescuing critical C9ORF72 ALS/FTD gain- and loss-of-function disease processes, respectively.
Before and after 2009, all major press groups in Fujian started to establish e-newspapers and transfer the newspaper contents to their official news websites to cope with the loss of readers as a result of the prevalence of computer technology as well as to maintain and expand the influence of traditional media. The popularity of Weibo and WeChat emerged between 2011 and 2012, thereby prompting the local press groups in Fujian to try out both platforms. During this period, newspapers were the pillar of the press groups’ revenue. Within most press groups in Fujian, Weibo and WeChat remained under the operation of newspaper editors and journalists on a part-time basis and received limited emphasis from the leadership.
To identify shared disease mechanisms between C9ORF72 and sporadic ALS motor neurons, we performed phenotypic analysis of patient iMNs. Previous studies have shown that reducing C9ORF72 protein levels using siRNAs impairs autophagosome formation in neurons (16, 17). To determine if the reduction in C9ORF72 protein levels caused by the C9ORF72 repeat expansion (4) is sufficient to block autophagosome formation in motor neurons, we expressed an mRFP-GFP-LC3 construct in iMNs to enable quantification of autophagosomes, which are GFP+ and mRFP+, whereas autolysosomes are only mRFP+ due to the lability of GFP at the acidic pH of lysosomes (18). Treatment with 50 nM bafilomycin to block autophagosome-lysosome fusion significantly increased the number of GFP+mRFP+ vesicles, indicating that this system could accurately monitor autophagosome number in iMNs (Figure 2A [CTRL, –bafilomycin vs. CTRL, +bafilomycin] and Supplemental Figure 2A; 3 controls). C9ORF72 ALS iMNs contained significantly fewer GFP+mRFP+ vesicles than controls (Figure 2, A and B [CTRL, inactive 3K3A-APC, +bafilomycin vs. C9-ALS, inactive 3K3A-APC, +bafilomycin]; 3 controls and 3 C9ORF72 ALS patients), indicating that autophagosome formation was impaired in C9ORF72 ALS iMNs.

Consistent with our previous studies (4, 8), a 12-hour pulse treatment of 10 μM glutamate induced significantly faster degeneration of C9ORF72 patient iMNs compared with controls (Figure 1, B [individual lines shown] and C [iMNs from all control or patient lines combined into a single trace for clarity], and Supplemental Figure 1, J and K). Glutamate treatment was not strictly required to elicit this effect, as withdrawal of neurotrophic factor supplementation also selectively induced the rapid degeneration of C9ORF72 patient iMNs without glutamate treatment (Figure 1D and Supplemental Figure 1L). Thus, ALS-causing mutations can cause iMNs to degenerate significantly faster than those from controls.

The impetus for junior journalists to participate in the converging news production is the same consideration that prevents senior journalists from collaborating with the new media centre, that is, to achieve the balance between gain and cost. The industry and the journalists ‘inside the system’ (tizhinei) are likely to opt for political safety over financial return, whereas their counterparts ‘outside the system’ (tizhiwai) ascribe considerable importance to the latter than the former.

The Chinese media have undergone commercial liberalization during the reform era. Interviews with media practitioners reveal that media reform has brought about three different types of newspapers that differ with respect to their degree of commercial liberalization. Based on a natural experiment during the anti-Japanese protests in Beijing in 2005, this article shows that urban residents found more strongly commercialized newspapers more persuasive than less commercialized newspapers. Provided that the state can enforce press restrictions when needed, commercial liberalization promotes the ability of the state to influence public opinion through the means of the news media.
Protein quantity was measured by the BCA assay (Pierce) and the proteins were run in a 10% SDS gel at 4°C, and the proteins were transferred onto an Immobilon membrane (Millipore). The membrane was blocked with 5% milk in 0.1% PBS-T (Sigma-Aldrich), incubated with primary antibodies overnight at 4°C, washed 3 times with 0.1% PBS-T, and then incubated with horseradish peroxidase–conjugated (HRP-conjugated) secondary antibody (Santa Cruz Biotechnology). After 3 washes with 0.1% PBS-T, blots were visualized using an Amersham ECL Western Blotting Detection Kit (GE) or the SuperSignal West Femto Maximum Sensitivity Substrate (Thermo Fisher Scientific) and developed on X-ray film (Genesee). The following primary antibodies were used: mouse anti-GAPDH (Santa Cruz Biotechnology, catalog sc-32233, 1:1000), mouse anti-NR1 (Novus, catalog NB300118, 1:1000), mouse anti–transferrin receptor (Thermo Fisher Scientific, catalog 136800, 1:1000), mouse anti-TUJ1 (Biolegend, catalog MMS-435P, 1:2000), rabbit anti-poly(GR) (Proteintech, catalog 23978-1-AP, 1:1000), anti-mouse–HRP (Cell Signaling Technology, catalog 7076S, 1:5000), anti-rabbit–HRP (Cell Signaling Technology, catalog 7074S, 1:5000), and anti-LC3 (VWR, catalog 101732-348, 1:1000).
From a new perspective, this article proposes four principles in understanding and interpreting Han dynasty brick and stone pictorial reliefs. First, adopt a three-dimensional viewpoint to describe and interpret the motifs. Second, follow the order of “bottom to top and right to left” in viewing the pictorial presentations. Third, take a holistic approach in appreciation, examining the pictorial reliefs together with the architectures of the tombs, memorial arches and other monumental pieces. And fourth, develop a comprehensive method of investigation, situating the Han pictorial reliefs in the historical context, including social customs, philosophical ideas, intellectual culture, economic development and advances in architecture. In writing the article, the author has done a systematic analysis, using contemporary historical texts, archaeological findings and modern works.

To answer the research question 1, the authors analysed the attitude of local journalists of Fujian towards new media with the method of survey. Available studies (cf. Wu and Zhang, 2015; Zhou, 2014) were used as bases to categorise the measurement of journalists’ attitude into two dimensions: The perception on the changes that new media have brought to news production and the overall appraisal of these changes. Measurement of the perception involves the following six items: (1) New media expanded the sources of news, (2) New media facilitated contact with work-related groups, (3) New media deepened knowledge on audience, (4) New media enhanced the requirements for journalists’ expertise, (5) New media intensified competitions with journalistic peers and (6) New media marginalised news gathering and editing inside the press group. A five-level scale was adopted (ranging from 1 to 5; 1 represents ‘strongly disagree’ and 5 represents ‘strongly agree’). The overall appraisal of those changes was measured by one item: What do you think of the overall influence of new media on news production? A five-point scoring was likewise adopted (1 representing ‘extremely negative’ and 5 representing ‘extremely positive’). Thereafter, demographic factors were measured, which included age, gender, educational degree, years at work and type of newspaper. At the end of the questionnaire, the respondents were asked if they are willing to accept an interview about their opinions on media convergence and leave their contact information.
Taking advantage of Fuchai's expedition to his north to defeat Qi, Goujian led his army and successfully attacked the Wu capital, killing the Wu crown prince, You. In the 24th year of his reign (473 BC), Goujian led another expedition against Wu, laying siege to the capital for three years before it fell. When a surrender from Fuchai was refused, Fuchai committed suicide and Wu was annexed by Yue. After his victory, Goujian ruthlessly killed Fuchai's scholars, even those who helped him (including Bo Pi), not allowing himself to make the same mistake Fuchai had made by sparing the lives of his enemies. However, Goujian would not stop there; he would later force Wen Zhong to commit suicide; Fan Li, knowing that Goujian was a man who can share woe but not wealth together, left Goujian after the defeat of Wu.

Meanwhile, partisans of the Donglin Academy faction, which had been devastated under Wei Zhongxian's influence, established political organizations throughout the Jiangnan region.[5] Chief among these was the Fushe, or Restoration Society, whose members were a new generation of scholars who identified with the old Donglin faction.[6] They succeeded in placing their members into high government posts through the imperial examinations of 1630 and 1631. The reversal of Wei Zhongxian's fortunes resulted in a renewal of the Donglin faction's influence at court, arousing great suspicion from the Chongzhen Emperor.[7] The nomination of Donglin favorite Qian Qianyi for the post of Grand Secretary led to accusations of corruption and factionalism by his rival Wen Tiren. Qian Qianyi was imprisoned on the emperor's orders. Though he was soon released, his status was reduced to that of a commoner and he returned to Jiangnan. Wen Tiren would later become Grand Secretary himself.[8]

After 2008, ‘full-media’ became a keyword with which scholars discussed China’s press industry. Studies that focus on the effects of full-media are categorised into two approaches: media economics and media performance approaches. The media economics approach generalises the experience and lessons of convergence practices and evaluates the influence of media convergence on newspaper development mostly from a business operation perspective by adopting case studies combined with the personal observation of researchers (cf. Tang and Cao, 2015; Yang, 2013). By contrast, the media performance approach analyses the method of content delivery on the new media platforms of newspapers as well as predicts (through content analysis) the influence of convergence on the progress of society’s democratisation from the media ethics perspective (cf. Huang, 2013; Cai, 2011).

Moreover, the major criteria of the present press industry in Fujian regarding job performance of new media editors involve the read count of postings and the number of fans. From the perspective of journalists, behaviour that editors repeatedly demand from the frontline journalists includes nothing more than ensuring the timeliness of press release. Their compliance is somehow related to the job performance evaluation of the editors themselves, thereby resulting in the additional decline in the willingness for coordination.

In light of the relationship between newspapers and websites, Mai (2012: 118–119) classified the path of media convergence of Chinese press industry into three types: extendedly ameliorated, establishing a convergence platform with newspapers as the core without changing the mechanism of content production and the structure of press industry; new media driven, setting up a convergence platform with new media as the core to push forward newspapers to progressively transform the flow, relationship and concept of news production; and fully transformed, replacing the original structure of the press industry with a brand new structure. Of the three, the ‘extendedly ameliorated’ approach has become the dominant path of media convergence because it rarely challenges the intrinsic structural restriction of press groups.

Despite the party-market corporatism, Lee et al. (2007) explained that significant multiplicity continues to exist in the developmental path of local media because of differences in power structure and market maturity. Guangzhou media represent the ‘market competition within party-state ideological limits’ pattern. For example, three press groups in Guangzhou compete fiercely with separate content strategies, while Guangzhou Daily and Yangchen Evening Daily cater primarily to daily life relevance and avoid ‘grand narratives’, Nanfang City Daily and Nanfang Weekend often expose official wrongdoings and advocate liberal ideas. Beijing media that are hierarchically structured in parallel to national, ministerial and municipal levels of administrative authorities compete both horizontally and vertically for power and market on behalf of their patrons and constituencies, thus represent the ‘managed diversity through a precarious of the emerging interest politics among counterbalancing power bases’ pattern. For example, as the authors put it, the supreme leader Mao had to turn to the Shanghai press to wage wars against the Beijing Daily on the eve of the period of the Cultural Revolution because his influence at that point could not fully reach the constituency of the Beijing Daily (Lee et al., 2007).

First, motor neurons derived from 6 sporadic ALS patients selected without bias toward certain genetic mutations or clinical symptoms display defects in autophagosome formation and accumulate NMDA receptors on their neurites. Both C9ORF72 and sporadic ALS iMNs exhibit these phenotypes, revealing an important area of mechanistic convergence between C9ORF72 ALS and at least some forms of sporadic ALS.

By the end of 2012, the circulation and advertising revenues of the Fujian press industry declined substantially, indicating its entry into a ‘cold winter’. Secretary-General Xi Jinping delivered the ‘8•19’ speech in 2014. Subsequently, Fujian’s press groups began to receive local fiscal support, actually ‘reaching a consensus’ and advancing media convergence. In 2014 alone, the Xiamen Daily Group gained substantial support from the municipal government, thereby pioneering the establishment of a ‘central control platform of converged media’ and a new media centre, which has been followed by several other press groups in Fujian.
To examine the effects of media convergence on news production, both survey and in-depth interview are adopted with the press industry of Fujian Province as the case of analysis. At present, four large-scale press groups exist in Fujian Province: Xiamen Daily Group, which owns three comprehensive daily newspapers (Xiamen Daily, Xiamen Evening News and Haixi Morning Post); Fujian Daily News Press, which owns three comprehensive daily newspapers (Fujian Daily, Strait News and Strait Herald); Fuzhou Daily Media Press, which owns two comprehensive daily newspapers (Fuzhou Daily and Fuzhou Evening News) and Quanzhou Evening News Press, which owns two comprehensive daily newspapers (Quanzhou Evening News and Southeast Morning Post). Overall, the size and influence of the Fujian press industry are ranked in the middle nationwide.

We and others have shown that iPSC-based models can recapitulate ALS disease processes, provided the iPSC donors harbor ALS-causing genetic variants (4–6, 8–12). As a key example, we have shown that iMNs from control lines consistently survive longer than iMNs from C9ORF72 ALS lines (n = 4 controls and n = 6 C9ORF72 ALS patients) (4, 5, 8). Because all environmental factors are held constant among the different lines in the iMN survival assay, we reasoned that sporadic ALS patients whose iMNs degenerate significantly faster than control iMNs likely harbored genetic mutations that promoted their observed motor neuron disease. Therefore, these sporadic ALS iMNs could serve as models of sporadic ALS.
