Interestingly, in the presence of 50 nM bafilomycin, sporadic ALS iMNs also contained significantly fewer GFP+mRFP+ vesicles than controls (Figure 2, A and C, and Supplemental Figure 2B; 3 controls and 5 sporadic ALS patients), indicating that autophagosome formation was impaired in sporadic ALS iMNs. Thus, both the C9ORF72 and sporadic ALS iMNs display decreased rates of autophagosome formation.

In 2006, the General Administration of Press and Publication of China began enforcing the first batch of the ‘China Digital Newspaper Laboratory Programs’, in which 18 national and local press groups were involved in officially unfolding the prelude of the ‘full-media convergence’, namely an intensive integration of new media into the system of traditional press. Since 2012, Chinese press industry has stepped into a ‘cold winter’. Throughout 2014 alone, the total revenue of newspaper circulation experienced a substantial decrease of 25%, with advertising revenue simultaneously decreasing by 15% (Cui and He, 2015). Several scholars suggested that vulnerable profit-making pattern, global economic recession and the decelerating growth rate of the domestic economy were the key factors for the predicament that Chinese press industry is experiencing (Zhao, 2015). However, the industry tends to ascribe the dramatic revenue decline to the prosperity of new media (cf. Cao, 2010; Zhou, 2015).

One of the goals of media convergence in the reconstruction process is to modify the journalist–editor relationship by veering from ‘journalist-centred’ to ‘editor-centred’, such that journalists shall mainly undertake the role of information gatherer, whereas editors perform the role of information synthesiser. Regarding real news production, most new media centre editors are recruited from the members of society who are familiar with new media but inexperienced in news gathering and editing; thus, they are mere ‘stevedores’ rather than ‘processors’ of texts. Given that editors lacked the required qualifications, journalists were forced to undertake additional tasks (e.g., verifying information and converting statements from frontline journalists into text) that should have been performed by editors.

Because autophagy induction can enhance TDP-43 turnover in motor neurons (26), we wondered whether 3K3A-APC treatment could prevent the cytoplasmic accumulation of TDP-43 in ALS iMNs. 3K3A-APC treatment for 6 days significantly increased the nuclear-to-cytoplasmic ratio of TDP-43 in C9ORF72 and sporadic ALS iMNs to control levels (Figure 3, G–J; 2 controls, 2 C9ORF72 ALS, and 6 sporadic ALS patients). Thus, 3K3A-APC treatment can rescue autophagosome formation, lower DPR levels in C9ORF72 ALS iMNs and rescue DPR-mediated neurotoxicity, and reverse the cytosolic accumulation of TDP-43 in C9ORF72 and sporadic ALS patient iMNs.

Jan. 2019, Check out the new paper: ‘‘Learning Nonlinear Mixtures: Identifiability and Algorithm’’. In this work we push forward parameter identifiability of linear mixture models (LMM) to nonlinear ones. LMM finds many applications in blind source separation-related problems, e.g., hyperspectral unmixing and topic mining. In practice, however, the mixing process is hardly linear. This work studies a fundamental question: if there is nonlinearity imposed upon an LMM, can we still identify the underlying parameters of interest? The interesting observation of our work is that: under some conditions, nonlinearity can be effectively removed and the problem will boil down to an LMM identification problem — for which we have tons of tools to handle.

Prompts from new media centre editors that ‘users responded intensely’ are often believed as too trivial to be ‘worth writing about’, hence are ultimately ignored by journalists. Editors blame the journalists’ reaction on lack of awareness, that is, journalists lack knowledge of new media, underestimate the intensity of competition between the press industry and commercial new media and overlook user demands. In fact, journalists expressed their concern regarding increasingly intense market competition in both the survey and in-depth interviews. They sensed that when various types of newspapers obtain news clues from new media and as reportage becomes increasingly homogenised, newspapers are confronted with increasingly intense competition.

When the Tianqi Emperor died in October 1627, he had no surviving heir (his last son died a year prior during the mysterious Wanggongchang Explosion). As the emperor's brother, Zhu Youjian, then about 16 years old, ascended the throne as the Chongzhen Emperor.[2] His succession was helped by Empress Zhang (widow of the Tianqi Emperor), despite the manoeuvres of the chief eunuch, Wei Zhongxian, who wanted to continue to dominate the imperial court.[3] From the beginning of his rule, the Chongzhen Emperor did his best to stem the decline of the Ming dynasty. His efforts at reform focused on the top ranks of the civil and military establishment. However, years of internal corruption and an empty treasury made it almost impossible to find capable ministers to fill important government posts. The emperor also tended to be suspicious of his subordinates, executing dozens of field commanders, including general Yuan Chonghuan, who had directed the defence of the northern frontier against the Manchu (later known as the Qing dynasty). The Chongzhen Emperor's reign was marked by his fear of factionalism among his officials, which had been a serious issue during the reign of the Tianqi Emperor. Soon after his brother's death, the Chongzhen Emperor immediately eliminated Wei Zhongxian and Madam Ke, as well as other officials thought to be involved in the "Wei-Ke conspiracy".[4]

For the DPR experiments, inactive 3K3A-APC was used as a control for any artifacts introduced into the downstream immunostaining measurements. After injection, the needle was left in place for an additional 8 minutes after the injection. Animals were euthanized 48 hours later. For injury size determination, brains were quickly removed, frozen on dry ice, and stored at –80°C until processing. Coronal sections (30 μm thick) were prepared using a cryostat, and every fifth section 1 mm anterior and posterior to the site of injection was stained with cresyl violet. The lesion area was identified by the loss of staining, measured by ImageJ and integrated to obtain the volume of injury.

To date, the pioneers of media convergence have been thoroughly studied by Chinese scholars. Other press groups as ‘followers’, particularly the local press groups that adopted the dominant path of media convergence, are less highlighted and rarely focused within journalism studies, although they are precisely what have crucially endowed the landscape of Chinese media convergence with regional diversity. The deficiency in relevant studies has entailed the urgent choice of Chinese scholars to focus on media convergence of the local press industry.
