Wang Xizhi is particularly remembered for one of his hobbies, that of rearing geese. Legend has it that he learned that the key to how to turn his wrist whilst writing was to observe how geese moved their necks. There is a small porcelain cup depicting Wang Xizhi "walking geese" in the China Gallery of the Asian Civilisations Museum in Singapore. The other side of the cup depicts a scholar "taking a zither to a friend".
A doxycycline-inducible (Dox-inducible) NGN2, ISL1, LHX3 (NIL) polycistronic construct was previously integrated into the AAVS1 safe harbor locus of the C9-ALS patient iPSC line using CRISPR/Cas9 editing (4). Dox-NIL iMNs were generated by plating at approximately 25% confluence on Matrigel-coated plates and adding 1 mg/mL Dox in N3 media plus 7.5 μM RepSox 1 day after plating. Mouse primary mixed glia were added to the cultures on day 6, and Dox was maintained throughout conversion. iMN cultures were harvested on day 26.

Upon the death of Yunchang and the accession of Goujian, Helü seized the opportunity and launched an attack on Yue. At the Battle of Zuili (槜李之战), however, Yue defeated Wu, and King Helü was mortally wounded. Before his death, he instructed his son, the later King Fuchai of Wu, "Never forget Yue!" Yue would be defeated three years later by a resurgent Wu, and Goujian captured, to serve as Fuchai's servant for three years before he was eventually allowed to return to his native state.

iPSCs were tested for mycoplasma before, during, and after the study and were negative. iPSCs were first differentiated into fibroblast-like cells to enable efficient retroviral transduction. iPSCs (1 × 106 cells/flask) were seeded in a T75 flask that was coated with Matrigel (Corning) and cultured in mTeSR until reaching 70% to 80% confluence. Cells were then cultured in fibroblast medium (20% FBS in DMEM) for 14 days. Cells were passaged 1:1 with Accutase (Innovative Cell Technologies) to another flask that was coated with Matrigel and cultured in fibroblast medium until they reached 90% confluence. Cells were then passed with 0.25% trypsin to 3 flasks that were coated with 0.1% gelatin and cultured in 10% FBS in DMEM. Reprogramming of the fibroblast-like cells was performed in 96-well plates (8 × 103 cells/well) or 13-mm plastic coverslips (3.2 × 104 cells/coverslip) that had been sequentially coated with gelatin (0.1%, 1 hour) and laminin (2 to 4 hours) at room temperature. Seven iMN factors were added in 100 to 200 μL fibroblast medium per well of the 96-well plate with 5 mg/mL polybrene. Cultures were transduced with lentivirus encoding the Hb9::RFP reporter 48 hours after transduction with transcription factor–encoding retroviruses. On day 5, primary mouse cortical glial cells from P1 ICR pups were added to the transduced cultures in glia medium containing MEM (Life Technologies), 10% donor equine serum (HyClone), 20% glucose (Sigma-Aldrich), and 1% penicillin/streptomycin. On day 6, cultures were switched to N3 medium containing DMEM/F12 (Life Technologies), 2% FBS, 1% penicillin/streptomycin, N2 and B27 supplements (Life Technologies), 7.5 μM RepSox (Selleck), and 10 ng/mL each of GDNF, BDNF, and CNTF (R&D Systems). The iMN cultures were maintained in N3 medium, changed every other day, unless otherwise noted.

Through the 1630s, rebellion spread from Shaanxi to nearby Huguang and Henan. In 1641, Xiangyang fell to Zhang Xianzhong, and Luoyang to Li Zicheng. The next year, Li Zicheng captured Kaifeng.[12] The year after that, Zhang Xianzhong took Wuchang and established himself the ruler of his Xi kingdom.[10] Court officials offered a number of unrealistic proposals to stop the rebel armies, including the establishment of archery contests, the restoration of the weisuo military colony system, and the execution of disloyal peasants.[13] Li Zicheng took Xi'an in last 1643, renaming it Chang'an, which had been the city's name when it was the capital of the Tang dynasty. On the lunar New Year of 1644, he proclaimed himself king of the Shun dynasty and prepared to capture Beijing.[14]
